Methods: Primary cultures of normal and scleroderma skin fibroblasts, and fibroblasts from PPAR- null mouse embryos, were used. COL1A2 transcriptional activity was studied by transient transfection assays. Protein expression and phosphorylation were determined by Western analysis. Nuclear protein DNA binding activities were examined by gel shift and DNA affinity precipitation assays. Subcellular Smad distribution was examined by confocal microscopy.
