PPAR- is a broadly expressed nuclear receptor that functions as a ligand-activated transcription factor induced by prostaglandins, lipids, and antidiabetic drugs. In addition to its role in regulation of energy metabolism, PPAR- is increasingly implicated in inflammation and tissue repair. We previously showed that PPAR- blocked stimulation of collagen gene expression in skin fibroblasts. In the present study, we investigated the molecular mechanisms underlying PPAR- suppression of profibrotic TGF- responses. Methods: Primary cultures of normal and scleroderma skin fibroblasts, and fibroblasts from PPAR- null mouse embryos, were used. COL1A2 transcriptional activity was studied by transient transfection assays. Protein expression and phosphorylation were determined by Western analysis. Nuclear protein DNA binding activities were examined by gel shift and DNA affinity precipitation assays. Subcellular Smad distribution was examined by confocal microscopy. Hat is caregiving and how do you define a family caregiver? What is the common thread that ties together those of us who care for spouses, children, parents, siblings, partners or friends who are chronically ill, frail, or disabled?
